Induction of Shoot Promordia of Hibiscus L. ¢¥honghwarang¢¥ by Suspension Cultures

Abstract
This study was carried out to determine the optimum cultural condition and method for in vitro mass production of Hibiscus syriacus L. ¢¥Honghwarang¢¥. Callus was induced in MS medium supplemented with 2 §·/L 2,4-D in 8 weeks. Subsequent suspension culture for proliferation was conducted with MS liquid media supplemented with 2,4-D and TDZ, and callus growth and shoot primordia formation were examined after 4 weeks. Then suspension was transferred to hormone-free MS medium, and shoot primordia formation and shoot regeneration were examined after 4 weeks. During the first 4 weeks, treatment of 0.1 §·/L TDZ was the most effective on callus growth. Formation of shoot primordia was the best at the MS medium supplemented with 0.01 §·/L TDZ. When the suspensions cultured in each medium were transferred to hormone-free media, most shoot primordia were formed most from callus cultured at MS liquid media supplemented with TDZ 0.01 §·/L. Shoot primordia induced from culture at 0.1§·/L TDZ were regenerated to shoot. We obtained about 30 calli from only one shoot-tip, and subsequently 20 to 30 shoot primordia were formed from each callus.
